A scientific and innovative team engaged in biology/chemistry-pharmacy-medicine interdisciplinary research. The undergraduate/postgraduate educational backgrounds of the team members cover a wide range of disciplines (biological sciences, biotechnology, bioengineering, biochemistry and molecular biology, cell biology, microbiology, biomedical engineering, mechanics, food science and engineering, pharmaceutical engineering, environmental engineering, and microscale science and technology, etc.), which provides a foundation of talents for interdisciplinary research.

Pharmaceutics期刊的细胞囊泡专刊“Advances of membrane vesicles in drug delivery systems”第1版完美谢幕,第2版已启动,欢迎投稿!

      历时约一年,本人倾心尽力为Pharmaceutics期刊(IF2022=5.4)客座编辑的关于细胞囊泡药物载体的专刊“Advances of membrane vesicles in drug delivery systems”(https://www.mdpi.com/journal/pharmaceutics/special_issues/Vesicles),最后一篇文章终于于近期正式发表,为本专刊第1版完美谢幕:

 

* 该专刊先后共收到了六十多篇投稿,经严格的同行评审后,最终发表了26篇文章;

* 该专刊发表了来自中、美、英、意等约十个国家的优质文章,特别是邀请发表了“国际细胞外囊泡学会(ISEV)”年会主席的研究论文;

* 截至近日,该专刊页面已收获4万+的阅读量;

* 该专刊在2022年正式发表文章6篇(2023年发表20篇文章),目前已共获得27次引用(平均每篇文章4.5次引用)。

陈勇研究员课题组开发了一种原位分离细胞核及核蛋白的新方法(原始新闻来自于南昌大学高等研究院官网-后附链接)

      在许多生物学研究领域(包括细胞生物学、分子生物学、发育生物学等)以及疾病相关的研究中,都涉及到信号通路及其相关关键分子的确定和相互作用。通过信号通路,外部刺激(或信号)由细胞外传导到细胞质再进入细胞核,通过一系列分子(包括细胞核蛋白)来调节特定基因的表达。因此,细胞核蛋白的分离和定量是信号通路研究的最重要的方面之一。细胞核及核蛋白分离纯化的质量决定了下游研究(例如常进行的蛋白免疫印迹和蛋白质组学等)的质量。至今,分离细胞核或核蛋白的方法有很多,包括超声法、酸解法、酶解法、蔗糖法、去垢剂法等。其中,去垢剂法的使用较普遍,发展了成熟的商业试剂盒。

      南昌大学高等研究院陈勇研究员的课题组在其他课题的研究过程中偶然发现,贴壁生长的细胞,在特定条件下,其细胞质成分能够被某些试剂原位剔除,只剩下仍然留在基底上的细胞表面囊泡和细胞核成分。基于此偶然发现,课题组先后发展了三种新方法:(1)发展了一种分离细胞表面囊泡的方法,并验证了分离纯化的细胞表面囊泡作为一种新型纳米药物载体的可能性(于2020年已发表在纳米生物学领域的知名学术期刊Journal of Nanobiotechnology;Zhang Y. et al. J Nanobiotechnol. 2020. 18: 69; DOI: doi.org/10.1186/s12951-020-00625-2);(2)发展了一种原位暴露细胞核及原位测量细胞核力学性质(例如刚性)的新方法(于2021年已发表在细胞生物学领域的老牌主流学术期刊BBA-Molecular Cell Research;Wang K. et al. BBA-Mol Cell Res. 2021. 1868: 118985; DOI: doi.org/10.1016/j.bbamcr.2021.118985);(3)近期又发展了一种原位分离细胞核或核蛋白的新方法,并验证了该方法的有效性,相比传统试剂盒分离方法具有更高的产率和更低的细胞质蛋白污染。传统方法将贴壁细胞变成悬浮细胞后进行后续的细胞和细胞核破碎从而获得细胞核或核蛋白,很难将细胞质蛋白去除干净。新方法是在贴壁状态下原位剔除细胞质成分(甚至可以在贴壁状态下破碎细胞核获得核蛋白),效果相当于进行了一次柱分离,因而能够显著提高产率及减少细胞质蛋白杂质(简单原理如下图所示)。新方法具有非常大的应用潜力和应用市场。

      该研究工作的成果近日已正式发表在生物学综合性权威期刊Biological Research上(doi.org/10.1186/s40659-023-00429-2),且该研究方法已获得国家发明专利授权。博士研究生秦颖和已毕业硕士研究生周云为该论文的共同第一作者,陈勇研究员为论文的通讯作者,南昌大学为成果唯一署名单位。该研究工作得到了国家自然科学基金项目、江西省自然科学基金重点项目、南昌大学学科交叉创新基金重点项目的资助。

曹迁永/陈勇跨学科交叉合作团队 发展一种谷胱甘肽激活型聚集诱导发光聚合物光敏剂 可用于杀死肿瘤细胞(原始新闻来自于南昌大学高等研究院官网-后附链接)

      谷胱甘肽(GSH)是由谷氨酸、半胱氨酸和甘氨酸组成的含有-酰胺键和巯基的三肽,存在于几乎每一个人体细胞中。GSH在人体内浓度异常与一些疾病相关,如肿瘤细胞内的浓度能达到1-10 mM,远远高于在正常细胞内的浓度, GSH可作为区分肿瘤组织与正常组织的生物标志物。光动力疗法(PDT)是一种无创、低毒、高选择性的癌症治疗方法,其核心是光敏剂(PS)药物分子。传统的PS如卟啉和氟硼吡咯等衍生物已经在临床上应用,但存在聚集诱导淬灭(ACQ)效应,影响其活性氧产生效率及治疗效果。同时,目前报道的PS大都不能区分正常组织与肿瘤组织,对正常细胞在PDT过程中也会产生光毒性。因此,有必要发展具有更大潜力的基于肿瘤标记物如GSH激活的新型光敏剂。

      南昌大学化学与化工学院的曹迁永教授和南昌大学高等研究院的陈勇研究员组建的“化学-生物医学”跨学科交叉合作研究团队,在近十年开展了深入的跨学科合作研究,产生了一系列重要研究成果。近期,该团队针对上述存在的问题和需求,发展了一种基于GSH激活型聚集诱导发光聚合物光敏剂。该光敏剂以聚降冰片烯为主链,侧链分别负载四苯乙烯AIE型光敏剂及含二硫键二茂铁猝灭基团。该光敏剂由于二茂铁至AIE荧光团的光致电子转移(PET)过程荧光及1O2产生能力都很差;当GSH存在时,发生了侧链二硫键断裂反应抑制了上述PET过程,荧光及1O2产生效率得到恢复。该新型聚合物光敏剂纳米颗粒对细胞内GSH有较好的荧光开启响应,对GSH灵敏度达到28.65 nM,可用于区分正常细胞与癌细胞;同时在白光照射下有很很好的1O2产生效率,对肿癌细胞有很好的毒杀能力。该研究工作在光敏剂和肿瘤诊疗技术等领域的发展中都将具有重要意义。

      该跨学科合作研究成果近日正式发表在化学-分析化学专业的国际知名学术期刊Talanta上(DOI: 10.1016/j.talanta.2023.124473)。曹迁永教授和陈勇研究员为论文共同通讯作者,博士研究生稂炜和硕士研究生陈丽珍为共同第一作者,南昌大学为成果唯一署名单位。该研究工作得到了国家自然科学基金项目的资助。
